I am full Professor at University of Tours, France. Since 2012, my research focuses on designing and producing original antibody-drug conjugates (ADCs) for applications in oncology and beyond. Original ADCs technologies, developed with Dr Caroline Denevault, include new bioconjugation techniques, original drugs presenting unusual mechanisms of action, new release drug mechanisms and conjugation of new antibody formats.An antibody-drug conjugate (ADC) is a targeted therapy, allowing selective delivery of a cytotoxic agent, thereby limiting usual systemic toxicity. Fluorescent probes can also be grafted to antibody for imaging. But almost all ADC feature a heterogeneous molecule loading (various numbers, various locations). We propose to optimize ADC construction, using a judiciously designed linker, which allows attachment to a native mAb and controls regiospecific grafting. This should be applicable to all antibodies (IgG and formats).
