I work within the ANR HIRAQIM project, conducting research on Arabidopsis thaliana to study mutations affecting water transport within the plant and their impact on immune responses to the pathogens Xanthomonas campestris and Pseudomonas syringae. The goal is to deepen our understanding of how these mechanisms contribute to plant defense.

Lab : Institute of Plant Sciences of Montpellier (IPSiM)Team :Root System Development and Plasticity Study of Peptide Hormones and Their Role in the Regulation of Proteoid Root Development in White Lupin.This internship focuses on understanding how white lupin adapts its root system to low phosphate conditions by studying the role of peptide hormones (CLE and CEP) in the development of proteoid roots. Techniques include hydroponic culture, root phenotyping, and the development and 3D printing of split-root hydroponic systems or prototypes for innovative experiments.

UMR DIADE, team : Epigenetic Regulation and Seed Development (REGD)Update and Maintenance of Seed Stocks: Managed the sowing and monitoring of cultures for Arabidopsis plants, including mutants and reporters.Genotyping and Validation: Conducted molecular validation using PCR genotyping and cellular validation through microscope observations to ensure accurate identification of plant traits.Plasmid Production: Produced plasmids for the genetic transformation of Arabidopsis thaliana, contributing to the study of plant physiology and development.

UMR PHIM, team : Xanthomonas Plants Interactions (XPLAIN)Still in the continuity of the proffessional license internship, I work on the XoCo project, Co-infection of Xanthomonas.

UMR PHIM, Xanthomonas Plants Interactions team (XPLAIN)I focused on methodological developments for exploring rice leaf co-infections by bacterial pathogens. My responsibilities included bacterial transformation by electroporation, plasmid extraction, and various DNA extraction methods (PowerSoil/CTAB).I performed simple rice leaf inoculation and co-inoculation using infiltration, leaf clipping, and sponge techniques. For bacterial population quantification, I employed dilution plating and QPCR methods. Additionally, I prepared libraries and conducted nanopore sequencing of bacterial DNA extracted from rice leaves, including adaptive sampling tests for bacterial DNA enrichment.

Team : Post-Transcriptional Reprogramming of Gene Expression in Response to Stress (SIPTRIP)During my internship at the Genome and Plant Development Laboratory (LGDP), I focused on the development of transgenic lines and phenotyping mutants of Arabidopsis thaliana. My responsibilities included:Selection of Homozygous Transgenic Lines: Assisted in selecting homozygous transgenic lines of Arabidopsis thaliana by inserting genes encoding proteins and verifying their expression through Western blot analysis.In Vitro Culture Techniques: Gained expertise in preparing culture media, performing sterile seed sowing, and conducting in vitro plant culture.Phenotyping of Mutants: Conducted phenotypic analysis of Arabidopsis mutants under different growth conditions to identify stress response traits.Western Blot Analysis: Performed protein extraction and Western blotting to validate the expression levels and sizes of transgenic proteins.
