Honorary Research Associate
Whole-genome CRISPR/Cas12a screens potentially yield a large number of meaningful biological information. As an extension of my work in atypical teratoid/rhabdoid tumors (ATRT), I am currently focusing on data analysis where I delve into ATRT's whole-genome genetic dependencies (the output of CRISPR screens) to identify unique biological vulnerabilities of therapeutic potential.
Cancer Research Intern
Together with the Next Generation Precision Medicine Program at the Centre of Cancer Research, I explored novel therapeutic strategies for a yet devastating pediatric brain tumor (i.e. atypical teratoid/rhabdoid tumor (ATRT)) through robust functional genomics and pharmacological means. For the first time, the largest cohort of ATRT cell lines was subjected to whole-genome CRISPR/Cas12a screening and high-throughput drug assays.
Teaching Assistant Of Cellular And Clinical Pathology Laboratory
The Cellular and Clinical Pathology Laboratory is the first biomedicine-exclusive lab course for second-year students. The cellular pathology scheme introduces students to good practise of mammalian cell culture and maintenance, conducting treatments on cell cultures, as well as relevant assays (e.g. cell viability assay, mycoplasma contamination detection, karyotyping). Meanwhile, the clinical pathology scheme encompasses blood handling, various blood characterisation and quantification methods, blood pressure measurement, contamination detection on blood and urine, as well as complete urinalysis. My roles as a teaching assistant are helping lab preparations, briefing students on session-relevant theories, assisting students' lab practise and organising their observation schedules, grading assignments, and welcoming student consultations while monitoring their progress.
Tutor Of Molecular And Cellular Biology
Under close supervision by lecturers, I conducted weekly remote meetings with a group of second-year students to have thorough discussions and assign them with quizzes on the materials relevant to the Molecular and Cellular Biology course. During the discussions, I often simplified principles essential for students in understanding weekly materials and addressed as many queries as they had. This tutoring period was intended to cover the topics of:- Molecular biology: Transcriptional, post-transcriptional, translational, and post-translational regulations of gene expression- Cellular biology: cellular organisation and motility, cell division and cell death, membrane transport, cell signaling, cell-to-cell and cell-to-matrix interactions, and stem cells
Intern Of In Silico Crispr Designing Project
The in silico CRISPR designing project encompasses two collective projects—’the generation of stable cell lines’ and ‘isothermal amplification and CRISPR diagnostics’. The former project employs the CRISPR/Cas9 technology to establish knock–in (KI) strategies for the generation of cell lines with a stable expression of the gene of interest (GOI). I was responsible for devising a CRISPR/Cas9–mediated GABA A receptor beta-2 subunit (GABRB2) KI strategy at an in silico level with the help of Benchling, as part of the generation of a stable cell line expressing a complete GABA A receptor. My strategy highlights the transfection of two plasmids (i.e. pCas9(BB)-2A-Puro (PX459) V2.0 and pAAVS1-P-MCS), preceded by appropriate cloning, that would be sufficient to provide human cells with all components necessary for the KI system. The assembly could be implicated for future generation of a stable cell line expressing the GABA A receptors as an in vitro disease model.
Teaching Assistant Of General Microbiology Laboratory
The General Microbiology Laboratory course was intended to introduce first-year students to the aseptic technique, bacterial culture, microbial characterisation and quantification, as well as antibacterial resistance test. As a teaching assistant, I was involved in preparing experiments, briefing students on relevant theories, assisting students' lab practise, and grading lab journals and reports.
