I had worked as a junior research fellow at ICAR-Sugarcane Breeding Institute, Coimbatore under the extramural research project “Pyramiding of transcriptional factor and ROS candidate genes for improved drought tolerance in sugarcane”. Sugarcane is a highly water intensive crop with an average water requirement 1800 to 2200 mm. Drought is considered to be the most deleterious abiotic stress in sugarcane. In this project we had transformed three different genes involved in drought tolerance viz, APX, CAT and ROS through particle bombardment method of gene transfer. I had worked as a project assistant at the same institute under the DBT project “Deciphering interacting partners of PAMPs /Effectors of Colletotrichum falcatum that triggering innate immunity in sugarcane”. Red rot is one of the serious diseases in sugarcane. Pathogen-associated molecular patterns (PAMPs) are recognized by pattern-recognition receptors (PRRs), which play a key role in innate immunity in the recognition of pathogens. In this project, I was assigned the task of developing a protoplast transformation protocol in Colletotrichum falcatum. Standardisation of the protoplast transformation procedure was completed. The gene CfPDIP responsible for the pathogen virulence was mutated and inserted in to a vector and was eventually used for protoplast transformation.I had working experience in molecular techniques like DNA extraction, RNA extraction, PCR, protoplast isolation, Microbial and plant genetic transformation, Marker assisted selection etc.,
