The impact of this work lied in incorporating in vitro evolved sensors of exquisite specificity for their target protein biomarkers in completely genetically encoded biosensor designs. To this end, I successfully prototyped two designs; one design utilised SELEX derived RNA-aptamer sensors incorporated into mRNA constructs to engineer ligand regulated sensor devices that produced a reporter protein output in cell-free extract reactions, and the second design utilised antibody fragments in a whole-cell bio-sensing platform.
