These electrodes create electric fields to manipulate nanowires in solution beneath the nano-pen.• Recorded high-magnification video of nanowire motion under varying voltages to examine electric field effects on nanowire movement.Nano-Pen Manipulator (NPM) System Development:• Designed and built a custom linear rail system with FUYU™ rails and motors, integrated into an aluminum frame with an air-cooled power supply for Arduino-controlled stepper motors along x, y, and z axes.• Programmed automated homing routines and implemented real-time control and feedback using PySerial communication.NPM Video Analysis Software:• Developed a Python-based GUI with PyQt and OpenCV for real-time tracking and analysis of nanowire manipulation.• Enabled multithreading for parallel processing of video streams with filters such as thresholding, contrast adjustment, Gaussian blur, and background subtraction.• Integrated Kalman filtering for noise reduction, allowing smooth plotting of voltage versus velocity.Precision Calibration of NPM Linear Rail System:• Created a command-line tool for linear-rail calibration using template matching on recorded measurements.• Employed a multistage, multithreaded approach, first detecting in low-resolution areas and then refining positions with full-resolution searches, significantly improving computation time.• Used DBSCAN clustering to identify position clusters and Kalman filtering to highlight trends.• Developed z-axis calibration using the “Variance of Laplacian” for sharpness assessment, matching frames with reference images to estimate z-axis displacement.• Applied DBSCAN clustering on nanowire positions to estimate distances and used linear regression to calculate step-to-micron conversion, achieving an R² value of 0.9883.
