I joined the Ludington lab in its first year to develop a host-microbiome model system in order to answer fundamental questions regarding the formation of gut microbiomes, their maintenance along life, and what creates diversity in microbiomes between individual hosts.In parallel of setting up a germ-free and gnotobiotc fruit flies facility, I isolated, characterized, and genetically modified bacteria from wild and laboratory-reared Drosophila species (mainly Acetobacter and Lactobacillus species). I then developed a high-throughput feeding assay with which I individually fed thousands of germ-free fruit flies with precise and increasing doses of engineered microorganisms (bacteria expressing fluorescent reporters).With this setup, I brought insights on the stochastic processes sustaining gut bacterial colonization: different strains of the same species can have completely different colonization efficiencies within genetically identical individual hosts. I also found that, unlike in vitro, bacterial species promoting the colonization of others can show distinct colonization patterns inside the gut.Some of these findings can be found on Current Biology's website (https://www.cell.com/current-biology/abstract/S0960-9822(17)30570-5?code=cell-site), and more is to come soon on bacterial interactions within flies' guts...

RNA interference (RNAi) is an antiviral molecular defense triggered by virus-derived double-stranded (ds) RNA molecules, which are naturally generated in infected cells during viral replication. RNAi arms insects with a sequence-specific mechanism of controlling viral infection.My thesis work provides evidence that enterocytes, the major intestinal cells, are highly involved in dsRNA uptake in virus-infected insects. We hypothesize that the intestinal uptake of virus-derived dsRNA is the first step in mounting a systemic immune response against insect-borne viruses.
