As a Research Associate in the discovery team at MiNA Therapeutics, I conduct end-to-end high-throughput drug screening using primary and mammalian cell-based assays, cell transfection, and electroporation for RNA activation of target genes.Key Research Activities:Screen and Optimize: Identify and screen lead candidates, optimize chemistries, and fully characterize profiles, including potency, cytotoxicity, and cross-reactivity testing.RNAi (siRNA), CRISPR-Cas9, Cell Transfection, RT-qPCR, nucleic acid and protein extraction, gene expression characterization, and novel drug target identification.Assay Execution: Conduct mammalian cell-based assays for high-throughput selection and characterization of oligonucleotides, assessing potency, cytotoxicity, and mechanisms of action.Manage and conduct lead optimisation of HbF saRNA oligonucleotides, to aid the nomination of lead candidates for sickle cell disease. Included in vitro experiments in primary cells, CD34+ (hematopoietic progenitor cell) and cell lines H661s and K562s.Troubleshoot & Optimize: Resolve experimental challenges and optimize complex experimental designs for greater reproducibility and sensitivity.Data Analysis & Collaboration: Generate and analyze assay data, collaborating with Matrix R&D teams to present findings, meet project goals and refine protocols.Lab Operations: Manage and enhance daily lab operations, including sample tracking, solution preparation, and inventory control to ensure smooth workflows.

Modelling crosstalk between macrophages and Tumour cells in the Tumour-microenvironmentActivities and societies: During this project, we studied the genotypic and phenotypic characteristics of a novel cell line induced from THP-1 cells by exposure to a conditioned- medium from the Huh-7 cell line. After performing cell culture for these human cell lines, I genotyped the novel cell line by PCR technique and compared the DNA profile of the novel cell line to reference the STR locus. Then I performed Florescence-activated cell sorting (FACS) staining and Flow cytometry technique to analyse the expression rate of cell surface markers on these differentiated macrophages.
