Supervised by Dr. David W. ChristiansonHistone deacetylases (HDACs) are essential enzymes in the regulation of gene transcription. Their aberrantfunction has been shown to play a role in cancer and neurological disease. Typically, HDACs are responsiblefor the removal of acetyl groups from modified lysine side chains on a range of proteins. Removal of the acetylmoiety, allows the positively charged lysine side chain interacts with the negatively charged DNA and silencesgene transcription. Structural analyses of HDACs in complex with inhibitors, substrate, or substrate analogs, allows for the characterization of the molecular features. Structural studies are imperative to design selectiveHDAC inhibitors; which are being explored for cancer chemotherapy and mitigation of chemotherapy induced neuropathy
