Electrophysiology patch Clamp lab.PD Dr. Gerhard TrubePatch Clamp techniques developed (First patch lab at Roche) on primary cells cultures (neurons), and cell lines (N2a, HEK 293, COS, SF9) .I successfully developed a new fast application system including new design of valves and control units. A few months experience with the automated patch clamp Robots PatchLiner (Nation Technologies).Electrophysiology on Xenopus Laevis OocytesStarting with manual measuring the Oocytes, I successfully set up two measuring robots (RoboOcytes) from MCS ( Multi Channel Systems) that I have deeply improved over the years ( new power supplies, new measuring heads, etc.)When asked we could measure up to 100 compounds a week (with two machines)The last four years I was involved in the the constant improvement of the tests (writing new scripts for speeding up the measurements.)Teaching the method of the in-house preparation of the Oocytes and handling of the Xenopus frogs.Cell Culture Lab.With Mrs Veronique Graf* Primary cell cultures on all available formats at any concentrations (well plates , glass cover clips, culture chambers, electrodes arrays, etc.) for different labs including Patch-Clamp and biochemical analysis.* We developed a proprietary method for freezing and thawing the neurons (Cortex and Hippocampus)* We also developed a proprietary method for slicing the brain for patch clamp experiments. Slices of the P8 hippocampus were alive up to 6 months.* Culture and measurements on MEAs (multi array electrodes systems from MCS and Ayanda)up to more then 60 days from fresh dissociated embryonic tissue as well as from the frozen ones (hippocampus).I developed a new proprietary method for slices cultures on MEAs.* We developed all our culture and dissociating mediums, including numerous coatings and techniques.* We could also bring in culture adults neurons and keep them alive during a a few months.
