Project: unraveling the mechanism of resistance to Nasonovia ribis-nigri (lettuce aphid) in lettuce with a combined metabolomics, proteomics and transcriptomics approach. This was a collaboration with ENZA Seeds, KeyGene, Wageningen University, University of Amsterdam and the Academic Medical Center. I was responsible for the proteomics within this project. Furthermore, I initiated and coordinated the large-scale experiments and advised the partners on useful techniques and literature for the project.Tasks:- a method was developed to isolate, concentrate and purify phloem proteins; - Reducibility and quality of the method was checked with LC-MS; - 2-D electrophoreses and DIGE was optimised for phloem proteins;- an EST database was constructed in collaboration with KeyGene and LC-MS with phloem proteins was optimised in collaboration groups from the UvA and the AMC;-participation in laboratory management and supervision of students;-management of the stock control of chemicals and equipment and restock supplies and materials in accordance with established policies and procedures;-literature studies for journal clubs and presentation of the findings.

Projects: efficient targeted DNA integration in plants and the induction of somatic embryogenesis by protein delivery into plant cells.The spin-off company from the University of Leiden developed new technologies and innovated products in the field of gene and protein introduction in collaboration with DSM, KeyGene and the University of Leiden.Tasks:- creation of T-DNA constructs and stable transgenic A. thaliana lines for all the projects and detection of single T-DNA insertion lines with SB;- analysis of gene targeting events and results of PEG-mediated protoplast transformations with fluorescence microscopy;- generation of stabile transgenic lines and maintain different types of cell cultures.
