Case Award Phd Research Student In Life Sciences

This role involved the management of a 4-year project as part of a broader research team in the School of Life Sciences. I was responsible for the direction, planning and execution of complex experiments, the supervision and training of technicians and undergraduate students and the reporting of scientific data in a variety of formats to a wide range of audiences.My PhD research was centred around the biochemical characterisation, functional analysis and structural determination of small Cysteine Rich Proteins, believed to be involved in nutrient allocation during cereal seed development.My project involved developing bespoke analytical tools for the characterisation of an unknown maize gene through biochemical analysis. This included engineering, expression and purification of recombinant proteins together with the generation and optimisation of custom monoclonal and polyclonal antibodies. Using GUS and GFP fusion constructs and immunohistochemistry together with confocal, epifluorescent and optical microscopic analysis, a clear understanding of a novel expression pattern was built.Generation of transgenics and a variety of phenotypic analyses supported by statistical analysis determined the function of an entirely novel maize gene with a complex expression profile. Gene function was disrupted using RNAi technology and the knock-down phenotype was analysed by microscopic analysis, genetic complementation, qRT-PCR and RNAseq. This provided evidence for a role in developmental cell signalling, nutrient allocation and a function spanning the pre/post-fertilisation period.The protein structure was determined by in silico modelling and solution state NMR spectroscopy to the aim of determining a structure-function relationship. To achieve this, a library of structural chimeras was generated and expressed in Arabidopsis as part of a functional assay to determine any cross-species functionality of CRP structural components and highlight function critical sequences.

The overall aim of this project was to dissect the role that small cysteine-rich proteins (SCRs) play in maize transfer cell development and assess their impact on seed development. This was achieved by the biochemical, molecular and cell biology characterization of maize transgenic lines that miss-regulate the expression of these proteins (Gutierrez-Marcos et al., 2004) and by the analysis of mutants that affect transfer cell development. Overall, the project provided novel insights into the role that SCRs play in the regulation of seed development in plants
