Silvio GutkindThesis Project: Investigate the role of GPCR-Protein Kinase A (PKA) signaling axis in regulating interferon type I innate immunity in macrophages.• Conduct extensive signal transduction pathway analysis with various biochemical, molecular, and cell biology techniques to elucidate upstream and downstream signaling events.• Perform cloning, lentiviral-mediated CRISPR deletion, and stable cell line generation for multiple proteins of interests; conducted subcellular fractionation and Western blot/flow cytometry analysis.• Develop custom R scripts for quantitative data analysis and data visualization.• Perform qPCR, RNA sequencing, differential expression, and GSEA analysis using R to characterize transcriptional signature.• Implement advanced imaging methodologies (confocal, electron, and lattice light sheet) and developed analysis pipeline to analyze intracellular trafficking events.• Lead inter-laboratory collaborations and mentor undergraduate students.

Advisor: Dr. Hua BaiProject: Study the role of SCAF1 in mitochondrial homeostasis upon peroxisomal stress.• Established a protocol to isolate mitochondria from cells and measured mitochondria functional assay.• Performed cells maintenance, molecular cloning, western blot, and confocal microscopy. • Designed genetic screening to identify candidate genes that mediates the communication between peroxisome and mitochondria.• Established an imaging protocol to measure mitochondrial membrane potential in fruit fly live tissues.

I am currently an undergraduate research assistant in the lab of Dr. Hua Bai. My independent project is to study the communication between peroxisome and mitochondria, and its implications on the development of cardiac diseases. I am responsible for experiment design, completion, and data analysis. Throughout this experience, I have the chance to gain experience on mammalian cell cultures maintenance, immunostaining, and confocal microscopy.

I was an undergraduate research assistant in the lab of Dr. Michael Cho. Projects that I am involved with is developing a potential candidate for HIV-1 vaccine utilizing structure-based approach. I am responsible for experiment set up, completion, and data analysis. Throughout this experience, I have the chance to gain experience on molecular cloning and protein purification techniques.

• Selected as one of 17 scholars from across the United States. • Participated remotely in the 4-week online training program including faculty research seminars, career-building workshops, and weekly journal clubs.• Facilitated a journal club in a group of 5 other interns and 4 Sloan Kettering graduate students.

Advisor: Dr. Jie WuProject: Identify KIF5B-RET mutants resistant to protein Tyrosine Kinase Inhibitor (LOXO-292)• Selected as one of 11 scholars from across the United States.• Established and incubated Ba/F3 cell cultures expressing KIF5B-RET mutants with LOXO-292.• Participated in professional development workshops and science integrity seminars on a weekly basis.

Freshman Research Initiative is a program that encourages freshman to get hands-on experience in research. I worked in a laboratory group lead by Dr. Donald Sakaguchi and Dr. Elizabeth Sandquist to conduct research about stem cell applications in retinal regeneration after injury in zebrafish model. I practiced immunohistochemistry technique, experimental design, and data analysis. At the end of the program, I presented my results in the Freshman Research Initiative poster session.
