Improvement of metabolomics analytical workflow and implementation to LC-MS and GC-MS datasets. Integration of metabolomics and transcriptomics datasets.

Thesis: “Metabolic footprinting of fermented milks after ingestion by healthy subjects”.Supervisor: Guy Vergères, François Pralong.Contribution to clinical study design and production of the test products (selection of lactic acid bacteria strain).Contribute to the setting-up of a metabolomics platform at Agroscope: defining and implementing an experimental workflow for the large-scale analysis of biological samples and the processing of high-resolution LC-MS untargeted metabolomics datasets.- Sample preparation and analysis workflow: serum and food samples, liquid-liquid and solid phase extraction, use of quality controls, large-scale UHPLC-qTOF-MS analysis with data acquired over multiple analytical batches (Bruker Maxis 4G)- MS/MS fragmentation analysis- Data pre-processing using Bruker Compass DataAnalysis, Progenesis QI and R-based tools (XCMS, MetMSLine, writing R scripts for adducts identification and analytical drift correction).- Data processing pipeline for the selection of biomarkers (data filtering, univariate statistical analysis, multivariate statistical analysis using R and SIMCA), GC and LC-MS metabolomics datasets.

Brain sampling, cryostat slicing, immunohistochemistry and laser capture microdissection (nucleus accumbens and hypothalamus).
