My responsibilities includes:- Planting transgenic Brachypodium distachyon and maintain them in a growth chamber and greenhouse setting. (This includes fertilizing the plants every week, making sure the plants are getting enough water, when is it appropriate to collect tissue and seeds from the plants.)- Collecting and preparing the Brachypodium distachyon mutant plants tissue for genotyping. - Utilizing direct PCR and gel electrophoresis to verify if the BT mutant plants are homozygous with insertion only in our area of interest. (Screening out BT mutant plants with multiple insertions).- Adapting, modifying, and planning how to conduct new experimental procedures that is needed for the project. - Utilizing bioinformatics tools like Coge, Phytozome v10.1 and BLAST to analyze the genes of interest and observe if this gene is conserved in other species. - Help manage the experimental data for the project which includes photos of the BT mutant plants, genotyping results, and primer tables. - Reading manuals and figuring out how to calibrate complex equipment used in the lab which include the QuantiStudio Flex 6 qPCR machine.- Mentoring an intern and figuring out what he do on a day to day basis.- Assisting the intern in finishing his small project by the end of the internship, where he will present a scientific poster on what he did for his project.

My responsibilities includes:- Planting the Brachypodium distachyon T-DNA lines.- Collecting BT mutant plants tissues and seeds.- Genotype to screen for chimeric T-DNA lines using direct PCR and gel electrophoresis.- Determining average fragment size using DNA Chip gel electrophoresis for DNA libraries to make sure they can be sequenced.

My responsibilities include: - Supervising and supporting students when they are conducting experiments. (Experiments includes DNA extraction using heat shock method, making Luria broth media plates, plating E.coli bacteria to confirm if bacteria were transformed, and HIC chromotography to extract GFP protein from the cell lysate.)- Prepare handouts and reagents needed for class.- Answer questions about the protocol and concepts taught.- Assist in teaching the students how to use the centrifuge, pipettes, spectrophotometer, balance,vortexer, and other equipment used in class.
