Thesis Title: Characterization Of Beryllium As A Novel Agent To Study Cell Cycle Arrest And Cellular Senescence.◆ Conceived, designed, executed, and managed graduate dissertation project using gene expression studies, PCR and RT PCR, protein analysis, cell based assays to characterize the effects of Beryllium on p53 signaling and cancer cell proliferation. ◆ Managed research laboratory projects and databases meeting deadlines and improving reliability of data storage.◆ Work done resulted in 3 peer reviewed manuscriptsGene Expression Studies:◆ Investigated an up-regulated intronic sequence in beryllium treated glioblastoma cell lines using PCR, cloning and sequencing.◆ Used microarrays to evaluate changes in gene expression in human cells and validated results with real time PCR resulting in discovery of Cyclin E2 modulation by beryllium treatment.◆ Employed shRNA / siRNA-mediated gene knockdown methods producing p53 knock down variants of glioblastoma cell lines.Protein Analysis:◆ Performed qualitative and quantitative measurement of proteins involved in DNA damage and cell cycle regulation using western blotting, fluorescence microscopy and flow cytometry.Cell Based Assays:◆ Developed a semi-HTS cell proliferation method to survey the response of over 15 cancer cell lines to beryllium treatment reducing the time required for screening drug response.◆ Used cell based assays to screen for caspase activation, changes in DNA synthesis, and changes in cell proliferation in response to drug treatments.
