💼 𝐌𝐨𝐧 𝐏𝐫𝐨𝐟𝐢𝐥 🔬Ingénieur en biotechnologies, spécialisé dans le développement des bioprocédés USP & DSP (production et purification de protéines recombinantes) depuis la création de Coraliotech. Je suis ainsi en charge de l’ensemble des processus de production de nos actifs aux échelles R&D et pilotes, de la préparation des matières premières jusqu’au contrôle qualité de nos produits. Un poste complet qui me permet de développer des compétences biotechnologiques toutes aussi variées autour de la culture cellulaire en bioréacteur, des systèmes de filtration et de concentration, de la chromatographie liquide, mais également en génie génétique, en microbiologie et en biologie moléculaire. Évoluant dans un univers définitivement startup à l’incubateur MONACOTECH, et doté d’un esprit plutôt curieux et créatif, j’aime étancher ma soif de connaissance en sortant de mon domaine scientifique pour découvrir de nouvelles compétences plus informatiques, notamment via la création de visuels, supports de communication et sites web. This innate resistance is often explained by the incapacity of aminoglycosides to enter the enterococcal cell.Our preliminary results indicate that the aminoglycosides interact with enterococcal cell wall with an affinity one order of magnitude higher than with cell walls of Bacillus subtilis and Staphilococcus aureus. We hypothesize that this interaction may explain the innate resistance of Enterococci to aminoglycosides.Enterococcus faecalis is a very diverse genus with differing strains displaying a wide structural and chemical variety of secondary polymers on the cell wall.PROJECTWe want to test whether the variety of cell walls translates in differing innate resistances to aminoglycosides. We have obtained a dozen strains of Enterococcus faecalis from the collection of Pascale Serror (1/3 clinical isolates, 1/3 isolated as food contaminants, and 1/3 intestinal commensals). We have tested ten strains and identified a strain whose innate resistance to aminoglycosides : JH2-2.We are going to test whether these differences in innate resistance to aminoglycosides correlate with differences in the affinities of the aminoglycosides with the cell walls of these strains. 1) Purification of Enteroccal strains cell walls.2) Affinity measure of aminoglycosides on cell wall by microcalorimetric study [Not achieved] Show less
