Graduate Research AssistantIowa State University Sep 2014 - May 2021Ames, Iowa, Us• Studied interactions of cadherin cell adhesion molecules at the single-molecule level both on live cells as well as cell-free context using AFM-based force measurements.• Performed chemical surface modifications to immobilize biomolecules on AFM cantilevers and substrates. Employed confocal fluorescence imaging to estimate the surface density of molecules; optimized surface chemistry protocols to achieve single-molecule measurements.• Performed model fitting, parameter estimation, and statistical analysis on 1000s of AFM f-d data to characterize the behaviors of biomolecular interactions. Developed data analysis and visualization tools using Matlab and Python. • Discovered the mechanism by which cells regulate the structures of cadherin cell adhesion molecules. Found a novel, asymmetric mechanism in cadherin-cadherin interaction.• Acquired, processed, and analyzed time-lapse images of migrating cells to quantify the migration behavior. Designed a novel experimental approach to eliminate the ambiguities of traditional approaches.• Carried-out PIV analysis to map velocity flow fields of migrating cells. Studied coordination in cells' motion by analyzing spatial correlation of velocity vectors, generated heatmaps to visualize directionality, and analyzed the presence of vortices in motion.• Perfomed AFM nanoindentation and used model fitting to extract cells' Young's modulus.• Helped setting up the lab after relocation. Trained junior graduate students on various lab techniques, equipment operations, and safety protocols. Conducted troubleshooting and preventive maintenance of various lab equipments.• Implemented Finite Difference Time Domain (FDTD) algorithms in C to model propagation of EM wave in 1 and 2 dimensional dielectric materials.• Characterized the properties of transmitted, reflected, and refracted waves under various boundary conditions. Studied dispersion and standing wave formation in dielectric media in the presence of scatterers. -
