Ubiquitin Specific Protease 21 (USP21) and its effects under radiation therapy in renal clear cell carcinomas. The USP21 is known to resist the radiation thus prevailing the cancer cells, therefore a siRNA mediated transfection was used to silence the USP21 gene and to validate the knockdown western blot analysis was conducted, while the conducting the western blot DNA damage response was also observed from PARP, ATM, and PH2AX. Apart from the siRNA knockdown, cells were also treated with an USP21 inhibitor drug called as Bay 805. Finally to observe its effects under the radiation therapy, transfected cells were plated in 6-well plates, exposed to radiation and incubated for 14 days and after 14 days they were dyed to observe the colonies.
