I realized my PhD in the Laboratory of Microbial Pathogenesis, under the supervision of Prof. Stewart T. Cole and Prof. Andy Oates.Tuberculosis (TB) remains one major cause of death worldwide due to a single pathogenic agent, Mycobacterium tuberculosis, and the emergence of drug-resistant bacteria urges the need of new antibiotics and treatment strategies. The current protcols to evaluate drug or vaccine efficacy in models of infection are extremely long, expensive and tidious. To circumvent these drawbacks, we are investigating several models of tuberculosis infection with the aim to improve their application in drug discovery and development. For example, by implementing an in vivo optical imaging platform, one could evaluate the progression of the disease in mice infected with Mycobacterium tuberculosis and follow up the effect of a new treatment in real time. Alternatively, zebrafish proved to be excelent models to study tuberculosis. Experimental infection in zebrafish embryos with Mycobacterium marinum replicates the initial stages of human TB. The simplicity of this model makes it an affordable system to evaluate the efficacy of new anti-TB drugs and can be easily adapted to a high-throughput in vivo model for drug discovery. Those approaches aim to fasten the discovery of new treatments and vaccines against TB and follow the ethical recommendations of the 3R's policy.
