    In the embryonic brain of the mouse, apoptosis peaks during neurogenesis and the dying cells correspond to radial glial progenitors, and migrating neuroblasts. The high vulnerability of migrating neurons may be due to high energetic requirements and production of oxygen free radicals, which can damage DNA and create double strand breaks. Studies from our laboratory and others suggest that the DNA double strand break repair protein Ku70 may serve dual roles in nuclear DNA repair and cytoplasmic sequestration of Bax (pro-apoptotic factor). To further investigate these roles, we have restored cerebral cortical Ku70 in Ku70 knockout embryos by means of in utero electroporation and examined whether this manipulations rescues migrating neuroblasts from apoptosis. We targeted migrating neurons in the dorsal region of the cerebral cortex and then labeled the migrating neuroblasts for apoptosis and cell type specific markers using immunohistochemistry. Preliminary studies using a control Red Fluorescent Protein plasmid suggest that this approach will allow us to investigate the neuro-protective role of Ku70 in migrating neurons.

    Malignant brain tumors are one of the most aggressive forms of cancer. CD133 is a cell-surface glycoprotein that marks cancer stem cells in brain tumors; however no functions have yet been attributed to CD133. To determine the biological function of CD133, I used RNA interference to eliminate CD133 in cell culture models. I designed lentiviral short hairpin RNA to inhibit CD133 production. RNA and protein levels of CD133 were analyzed using RT-PCR and western blotting. The results reported that the CD133 mRNA and protein levels were reduced and protein production was inhibited. The vectors I constructed will be used to test the function of CD133 in in vivo transplantation models.

This work was further characterized by categorizing the transcriptome at three levels: (1) the central nervous system, (2) the elementary components of a simple behavior: the gill-withdrawal reflex—by analyzing sensory, motor, and serotonergic modulatory neurons, and (3) processes of individual neurons.

    MicroRNAs have been shown to be crucial to development of the mammalian nervous system. Specifically, the spatiotemporal control of mRNA translation has an important role in synaptic development and plasticity. I worked on isolating microRNAs from the rat mammalian brain with the goal of localizing them to specific regions of the brain. This project ended with showing that microRNA, miR- 134 (a brain-specific microRNA) is localized to the synapto-dendritic compartment of rat hippocampal neurons and negatively regulates the size of dendritic spines (post-synaptic sites of excitatory synaptic transmission).
