-Expressed protein in bacterial system and purified through AKTA pure FPLC system (Tag/untagged/IEX/SEC chromatography) Agilent HPLC-Purified native PDE6 (membrane protein) from bovine rod photoreceptor using AKTA FPLC purification (gel-filtration, ion-exchange/Mono-Q chromatography) -Performed chemical crosslinking of protein with/out the binding partners, validated the data by SDS-PAGE followed by in-gel digestion and Mass spectrometry; MS data analyzed by protein prospector-Constructed Homology model using Modeller/ Integrative Modeling Platform (IMP)-Performed NMR studies to confirm the topology of proteins
