University professor is generally considered one of the most satisfying occupations. Freedom in scheduling is often considered one of the perks. Yes, we can work any 80 hours of the week we wish. Organizational skills are critical as we balance periods of intense effort writing a grant, exam, or promotion letter, with lighter ones requiring idea generation and planning.Since my first research experiences, through the tense days as a graduate student, postdoc, and untenured assistant professor, I was motivated by a desire to learn or create something new. As my research interests evolved I became more interested in having a significant impact on human health by exploiting the broader expertise of my group members and collaborators in a team effort to identify new compounds with novel biological activity, to develop both synthetic and biosynthetic methods to access these compounds, and to fully explore their therapeutical potential. We are typically assigned to teach two specific courses per year at either the undergraduate and/or graduate level. The top students are stellar and challenging, the average student is engaged, and the weaker students still appreciate your effort to guide them through one of the most difficult courses at the University. Although we get little official credit, researchers have an additional teaching load, the mentorship of their graduate students, postdocs, and undergraduate researchers. It is actually this area that attracted me most to this job over one in the pharmaceutical industry.I recall seeing a resume of one university faculty member where the section entitled, “University and Departmental Service” had only one entry, "I have served on many departmental and university committees". This is true to all faculty. Through my time at Notre Dame I have tried to utilize additional department and university service positions to represent and advocate for the faculty and graduate students as we evolve and grow our research enterprise.

Professor Of Chemistry & BiochemistryUniversity Of Notre Dame Jul 1995 - PresentNotre Dame, In, UsOur group is interested in the exploring the potential of polyketide natural products as chemotherapeutic agents particularly directed against cancer. Towards this end our group's expertise includes synthetic chemistry, molecular modeling, and molecular biology/biochemistry. In recent years the we have completed the total synthesis of the complex natural products, epothilones A, B, C and D, myriaporones 1, 3 and 4, peloruside A, neopeltolide, tolypothrix polyethers, gephyronic acid, diospongins A and B, dactylolide, and zampanolide. Our analogue design strategy is focused towards the determination of the structural and conformational constraints of binding. Our group has demonstrated that the information gained from what we have termed conformation-activity relationships complements classic SAR with the goal of providing a detailed pharmacophore model and assist in the design of future chemotherapeutic agents. In addition to our successful total syntheses and medicinal chemistry efforts, our group has already contributed significantly to the field of organic synthesis through the development of a number of new synthetic methodologies that solved key problems in our targeted efforts.Another unique aspect of our analogue design strategy is the exploitation of biosynthetic enzymes called polyketide synthases. We are actively pursuing the isolation and characterization of polyketide synthase gene clusters responsible for the production of biologically active natural products. We have demonstrated the semi-synthetic production of epothilone natural products and analogues through the use of genetically engineered organisms thus alleviating any concerns about the high cost of total synthesis of compounds of this complexity. -

ResearcherResearcher At The University Of Notre Dame Mike And Josie Harper Cancer Research Institute Jun 2011 - PresentOur laboratory has identified a number of new chemical entities with significant anti-cancer activity and is working to evaluate these leads in pre-clinical animal models to determine their potential for clinical studies. The laboratory uses a combination of synthetic, computational, and molecular biological studies. We are particularly interested in the correlation between unique natural product structures and their corresponding biological activities.The research is significant because, despite the advent of combinatorial chemistry and high-throughput screening during the past decade, the number of new biological targets that have emerged from the pharmaceutical industry is remarkably small. Natural products are especially important because evolution has created these compounds to transport and modulate biological function. The detailed study of natural products with unknown or unique modes of action represents a paradigm with the ability to identify new biological targets.Our laboratory aims to generate new sources of chemical diversity based upon the modification of polyketide scaffolds and explore their chemotherapeutic potential.
