Mainly our lab was dealing with the abiotic stress in plants, which focuses on functional validation and characterization of stress tolerant genes. I have joined here in a DBT India funded project “Development and evaluation of salt and drought tolerant transgenic rice”. During this entire period I was part of various projects, involving different genes and gene families like CBS, NCX etc. I have sharpened my skills and increased the understanding in the field of Plant Biotechnology during this entire period. Some details of my work are as follow:1. I have started my work with rice cDNA library screening, on sodiumchloride to explore some salinity stress responsive genes.2. To confirm their role in abiotic stresses I have done qRT-PCR for such genes with cDNA of plantlets undergone different abiotic stresses.3. Cloning of genes from rice genome in TOPO TA vector and confirmed them with sequencing and BLAST software.4. Cloning of stress responsive genes in Plant transformation vector and transformation into agrobacterium.5. Agrobacterium mediated transformation of the genes into rice, tomatotobacco and Arabidopsis for their functional validation.6. Confirmation of transgenic by tissue PCR, Southern hybridization, Western hybridization and RT PCR.7. To carry out comparative study between control and transgenic plantat protein level I have performed 2D gel electrophoresis (DIGE).

I have pursued my M.Sc. Dissertation entitled “Cloning of MAP kinase kinase from OryzaSativa and Agrobacterium mediated transformation” under the supervision of Dr. Alok.K.Sinha. Here I was trained to be an independent researcher. I added more knowledge, skills and understanding of molecular biology. This short time encouraged me to think deep inside the plant development and physiology, mainly I was interested in abiotic stress.
