Anti-tumor activity of transition metal complexes with DNA is an area of drug discovery research that has recently gained momentum. My lab's most recent project investigated the reactivity of di-metal, platinum and porphyrin compounds with salmon sperm DNA and cloned plasmid DNA. The compounds studied were water soluble diruthenium complexes, cis-Diammineplatinum(II) dichloride [Cisplatin], and 5,10,15,20-Tetrakis(1-methyl-4-pyridinio)porphyrin tetra(p-toluenesulfonate), [TMPyP]. Di-metal and platinum antitumor activity has been attributed to their ability to react with nucleic acids by forming intra-strand crosslinks. Three binding modes are used to describe these interactions; outside binding, outside binding with stacking, and intercalation. The presence of these adducts with various forms of DNA were measured using UV-Vis spectroscopy, relative viscosity, and electrophoretic mobility shift assays (EMSA). Intra-strand crosslinks of di-metal complexes with DNA may result in changes in melting temperature profiles (Tm), relative viscosity times, and migration rates using gel electrophoresis. It is hoped the results of these experiments might indicate how these di-metal complexes bind with double-stranded DNA and ultimately lead to favorable, alternative anti-cancer therapeutic agents.
