About Robert Sackstein Md,Phd

My efforts as a basic scientist and clinician are intimately intermeshed. I am a basic science immunologist/biochemist/molecular biologist with clinical expertise in internal medicine/hematology/immunology and, in particular, in hematopoietic stem cell transplantation (HSCT). Accordingly, my bench research efforts aim to elucidate biologic processes critical to improving outcomes for patients undergoing stem cell transplantation, such as: (1) hematopoiesis, and other cell and tissue regeneration from stem cell-based therapeutics; (2) tissue-specific lymphocyte migration (including the immunobiology of lymphocyte migration in host defense and in pathologic reactions such as graft-versus-host disease); and (3) pathobiology of tumor cell proliferation and tumor metastasis. The common thread for all these efforts is to manipulate the biology of cellular trafficking, as it pertains to stem cell transplantation and tissue regeneration (‘regenerative medicine’), to host defense/inflammation, and to cancer growth and metastasis.

    My clinical expertise is in hematopoietic stem cell transplantation (HSCT) and throughout my career I have performed basic research with the hope of creating novel strategies to improve outcomes for my patients. My research interest is in the regulation of cellular trafficking as it relates to HSCT, with a focus on how hematopoietic stem cells traffic to marrow and how alloreactive and regulatory lymphocytes gain access to target tissues in graft-versus-host disease (GVHD). In particular, my efforts are focused on defining the molecular effectors of cell migration with intent to define the optimal method(s) to deliver cells systemically to sites where they are needed. This work fundamentally seeks to improve hematopoietic and immunologic recovery after HSCT and, more broadly, to enable regenerative therapeutics and adoptive immunotherapy. To this end, my laboratory has developed a platform technology called “Glycosyltransferase-Programmed Stereosubstitution” (GPS) to enforce surface sialyl Lewis X (sLeX) carbohydrate substitutions on relevant cells, thereby conferring binding to the selectin class of adhesion molecules.
