During this experience, I deepened my knowledge in molecular and cellular biology by researching the genetic influence of neuropathological diseases on subtle changes in the brain. I mastered cell culture and biochemical techniques for DNA, RNA and proteins. One of my challenging roles was to develop an in vitro assay by combining electrophysiology and high-resolution calcium imaging for screening genetic mutations. The outputs are complex data on subcellular calcium influx and the spontaneous fusion of synaptic vesicles. With a team of experts, we developed SICT to automatise the analysis of a complex dataset. Moreover, I: ⁍ Trained students and supervised their research projects. ⁍ Wrote scientific reports and SOPs.⁍ Managed multiple projects, related teams and meetings.⁍ Attended scientific conferences and presented my result with posters and orally.

Kleefstra syndrome is a form of intellectual disability, and it is linked to the lack of EHMT1, a histone methyltransferase. The neuroscience research project aims to examine particular epigenetic mechanisms which affect homeostatic plasticity, a specific form of synaptic plasticity. My Master thesis reports this study “Epigenetic control of EHMT1 chromatin remodeler in synaptic homeostasis”.My role was to assess the endogenous EHMT1/ EHMT2 protein levels before and after the induction of homeostatic plasticity by western blot and immunocytochemistry. Moreover, I : ⁍ Set-up, executed and distributed a protocol for immunoprecipitation using rat primary neuronal cultures. ⁍ Isolated RNA and performed qPCR. ⁍ Analysed data, prepared reports and presented outcomes to the lab members.⁍ Attended lab meetings. ⁍ Improved my scientific network by attending presentations from international speakers and collaborating with other departments.

This research project in Immunology focused on understanding the intracellular LCK-mediated signalling in T cell survival rate during TCR activation. I was involved in cloning LCK mutants, producing lentiviral particles in T239 cells and measuring with immunocytochemistry the expression of LCK mutants in primary and Jurkat lymphocytes. I analysed the fluorescence of T cells expressing LCK mutants after TCR activation by flow cytometry. Being part of an international research team contributed to deepen my interpersonal skills and my passion for science and research.

I reported in my Bachelor thesis this research project in neuroscience. It investigates the mechanism of α-synuclein aggregation, a process involved in synucleinopathies. I analysed by qPCR and western blot the effect of 5-S-Cysteinyl Dopamine on the expression level and aggregation of α-synuclein in SH-SY5Y cells.With this experience, I learned how:⁍ to handle laboratory equipment.⁍ to follow general instructions and procedures as provided. ⁍ to apply computer skills for research, data analysis and presenting scientific reports.⁍ to report directly to supervisors.
