I am working with Prof. Navin Varadarajan on multiple projects, including-Production of recombinant 2’3’-cGAMP from Escherichia coli• Designed complete upstream and downstream bioprocess for synthesis and purification of 2’3’-cGAMP, a potential new generation vaccine adjuvant.• Performed codon optimization using HTS-based methods and performed vector design and conjugation for protein expression.• Led bioprocess development from plasmid design, cloning and culturing (upstream) to and purification (downstream) processes. Performed bioprocess optimization resulting in significant yield improvement (~30%) over previous studies, resulting in patent application.• Performed intermediate characterization using RP-HPLC and LC-MS, as well as endotoxin testing for final product.Development of commensal bacterial strains for immunomodulatory response• Studied engraftment of engineered commensal bacterial strains producing immunomodulator in mouse model. • Collected biological samples (intestine) to study immune cell activation profiles in Flow cytometry setup.Development of intranasal vaccine for SARS-CoV-2• Designed, performed and analyzed experiments in molecular cloning, recombinant protein production (expiF/CHO-K1) and animal studies to study vaccine responses for a fusion antibody-based vaccine with mucosal delivery system to improve antigen presentation at site of infection in SARS-CoV-2.

Worked with Prof. Sarika Mehra and Dr. Vikas Chandrawanshi on the following project-Metabolic Flux Analysis (MFA) in CHO-K1 cells• Created a systems biology approach for studying metabolic network comprising major reaction pathways of cellular metabolites using growth and secretome data from Tunicamycin treated CHO cells.• Mapped both extra/intracellular reaction fluxes by bootstrapping and solving the overdetermined linear system of fluxes, showing variation between control and Tunicamycin treated CHO cells shows altered metabolism in particular pathways.
