About Ronald Henry

Since 1995 I have been working in the Biotechnology industry developing bulk separation and down stream purification processes for bench, pilot and manufacturing scales. I have developed processes for pharmaceuticals (Chiron/Novartis), industrial fine chemicals and biofuels using synthetic biology platform (Amyris Inc.). I have broad experience in scale-up and technology transfer of downstream processes to domestic and international manufacturing sites including Brazil, Canada, France and Spain. I am seeking opportunities to work for cutting edge companies in biopharma, biofuels and fine chemicals utilizing microbial and mammalian cell culture, and synthetic biology platforms.

Developed novel separation and recovery schemes for yeast and E.coli derived sesquiterpenes engineered on a synthetic biology platform.• Developed a novel, scalable aqueous extraction and precipitation method for separating Artemisinic Acid from yeast fermentation broth. I am co-inventor on a patent for this unique method. • Developed bench and scalable methods for separating Farnesene hydrocarbon from E.coli and yeast fermentation broths with >90% recovery yields. • Developed new assay and screening methods to predict downstream recovery potential in new strains.• Developed and implemented new in-process assays to monitor downstream recovery unit operations performance at pilot and manufacturing scales.• Designed experiments to characterize where desired products partition in process streams, from fermentation harvest to final crude product.• Designed and implemented experiments to determine mass balance yields across all downstream unit operations for both pilot and manufacturing scales.Developed methods for cleaning up and preparing various crude feedstocks for yeast fermentations. • Designed and implemented centrifugation, filtration and other methods for preparing raw cane and sorghum syrups and molasses.• Collaborated on a grant project with the U.S. Department of Energy to prepare and test various cellulosic derived feedstocks. These included hydrolysates of Loblolly Pine, Sorghum and wheat straw bagasse. I was involved in the scale-up and technology transfer of the downstream recovery process for the hydrocarbon Farnesene to multiple domestic and international manufacturing sites including Brazil, Canada, France and Spain. Recovery processes that I developed at the bench scale were successfully scaled up to fermentations as large as 300,000 L.
  • Chiron
    Senior Development Specialist
    Chiron 2000 - 2007
    Emeryville, California, Us
Utilized broad hands-on experience in the development of E.coli fermentation, protein recovery and isolation processes, protein folding, and supporting analytical techniques. * Developed a stable, post-refold precipitate intermediate that allowed de-coupling of refold and purification operations. This work explored protein precipitation using acids, a variety of salts and PEG.* Worked on the development of a high-density fermentation for proteins expressed in microbial hosts using a limited glucose feeding strategy to minimize cytotoxic acetate production. * Developed stable recovery processes for proteins expressed in microbial and mammalian hosts. Projects included: - Determining and optimizing process chemistry. - Determining optimal equipment and environmental operational parameters, including range finding and determination * Produced small-scale GMP manufacturing lots to support clinical trials of both licensed and early stage products. * Developed process and equipment improvements for a large-scale solvent extraction recovery utilizing a centrifugal three-phase separator.Involved in the Fermentation/Recovery scale-up, technology transfer and validation of several commercial and clinical products expressed in E. coli and Pichia. I Provided commercial manufacturing support for the licensed products PDGF, IL-2 and IFN-β. I worked on joint projects with Pfizer, Entremed, Alexion and Zymequest.
