About Rosie Irwin

I aim to further our understanding of the physical basis for the workings of the mind through biochemical and biophysical research. Many questions motivate my work - what makes up a thought? How does the chemistry of the brain and body create the human experience? Is there a viable way to answer these questions?     Working under the advisement of Dr. Julie Forman-Kay in the Program in Molecular Medicine, I seek to understand the role of biomolecular condensates in spatial and temporal organization at the synapse. More specifically, I am working to understand how these condensates regulate mRNA translation and degradation. With this work, we hope to describe how changes in these dynamic structures influences development and synaptic plasticity.

    For my MSc thesis project, I worked in the labs of Dr. Rina Rosenzweig and Dr. Hagen Hofmann, employing single molecule FRET, advanced NMR techniques, TEM, and aggregtaion-prevention assays to understand the mechanisms by which ATP-independent chaperones prevent tau protein aggregation as well as the structural basis for such activity. We focused on the Hsp40 chaperones DnaJA2 and DnaJB1 and compared their function to that of small heat-shock protein HspB1. We determined that the two Hsp40s have distinct tau interaction profiles, which lay the foundation for unique yet complementary aggregation prevention mechanisms. Our results demonstrated a clear link between the tau species binding capability of a chaperone and its mechanism of aggregation prevention.

    As a RA II, I continued to assist in crystallography experiments for proprietary crystallization projects as well as a project for the Seattle Structural Genomics Center for Infectious Disease (SSGCID). This work involved setting up crystallization experiments, harvesting crystals, collecting data, and helping solve and refine both proprietary and SSGCID structures. I became more independent in my SSGCID project by taking on responsibility at all stages of the pipeline, from designing crystallization experiments to solving structures. I also gained some experience in ligand-binding NMR and differential scanning fluorimetry experiments.
