About Sam Laurel Stephen
Viral genetic medicine has reached clinics now: the central theme of the first part of my research career was on the genotoxicity of these vectors. During my PhD, I developed methods to study recombination of adenoviral vectors in vitro and in vivo. During my first post doctoral research at UCL, I worked on biotoxicity of retro and lentiviral vectors before moving on to generating a clinical grade producer cell line to generate lentiviral vectors under academic GMP conditions.During my second postdocotral research at the University of Leeds, I moved from understanding viral vectors to using them in translational research. We established a BSL II facility for the generation of Adeno/retro/lentiviral vectors and investigated the role of scavenger receptors in atherosclerosis using these viral vectors to upregulate and knockdown gene expression. Another arm of viral genetic medicine is the use of virus-like particles as vaccines. Self-asembling hepatitis B capsids seem ideal candidates in this role. We generated the VLPs in E. coli and then used a host of biophysical and molecular biology methods to characterise them, in collaboration with an industrial partner for use in clinical trials. I also collaborated on projects involving VLPs/Replicons based on Hep B surface antigen, poliovirus, MS2, Lambda bacteriophage, FMDV, EV 71, HPV, Norovirus and Ebola virus.Currently, I am working with viral vectors and vaccines, generated under ISO9001 conditions in an industrial setting. The vectors are generated, analysed and all the upstream, analytics and downstream processes are carried out in-house. We work with small scale cultures as well as bioreactors. We are also working on innovative projects in optimising the production and application of viral genetic medicine.
