    Directed research investigating the nerve dependence of limb regeneration, specifically in the axolotl salamander. • In an effort to identify and characterize neurotrophic factors (NTF) responsible for supporting the proliferation of blastemal cells during the early phases of regeneration, transcriptional changes were analyzed using fluorescence in situ hybridization (FISH) and Click Chemistry, focusing on previously identified NTF candidates. • Conducted a series of proteomics experiments investigating genes associated with histological and transcriptional changes after injury, focusing on the dorsal root ganglia and spinal cord.• Designed a fixture to image regenerating limbs, and completed simulations of the regenerating limb bud. • Experimented with 3D printing, casting, and molding to design replacement microscope imaging tubes to have the refracting index of glass.

This method is to be used on the next batch of 800 tendons, saving the team approximately 2 minutes per sample.

    We are working to adapt the properties of cryogels in order to allow for more versatile use throughout the body and, with the addition of stem cells, create a wound closure treatment.• Conduct trials on prevention of dopamine oxidation with varying antioxidants.• Run mechanical testing of cryogels (tensile, compression, sheer) using an Instron Mechanical Tester.• Perform adhesion testing using ASTM Standards for Sheer Stress, Burst Pressure, and Wound Closure using porcine skin as a wound model. • Design cryogels using APS/TEMED co-initiation chemistry into various shapes and sizes that can be compressed to 90% of their original volume.• Future steps: conduct cell studies to ensure viability within the cyrogels (ELISA, media preparation, cell imaging).Publication: Rana, D., Colombani, T.
