• Determined the effects on morphologic maturation and differentiation / functional activation of over expression of the nuclear envelope proteins. o Lamin A (LMNA) and SUN 2 inserted into plasmids and transduced into novel hematopoietic cell lines.• Plasmid preps, restriction enzyme digests, • Developed optimized protocol for transducing two myeloid models. o ER-HOXb8-GMSCF and ER-HOXb8-SCF cells,. • This technique was then used to transduce small inhibitory RNAs against key transcription factors, including GABPα.• Investigation into the possible effects of Liver X Receptor (LXR) functions on the growth and differentiation of a myeloid model were determined using LXR agonists and antagonists. o These data were critical to our studies of the Lamin B receptor in mediating cholesterol biosynthesis (as published in Subramanian et al, 2012, see below).
