Sr. RaAnokion Apr 2021 - Jan 2024Cambridge, Massachusetts, United StatesAt Anokion, I work in the cell biology group to maintain and develop new cell lines for routine protein expression and cell-based assays. I often collaborate with other groups to design new proteins for both in vitro and in vivo studies. I design construct plasmid maps, devise a cloning strategy, and prepare DNA for transient protein expression in mammalian cells. I use PCR and restriction enzyme cloning methods in in this role. I also purify constructs on an AKTA purification system using protein A/G columns and characterize these constructs with Western blotting. While at Anokion, I have developed several stable cell-lines that express receptors of interest for cell-based assays. I run cell-based assays to compare binding affinity and internalization of different molecules. I have experience with assaying primary cells and splenocytes in ex vivo studies as well. I am comfortable with FlowJo and CytExpert. I also have experience with multi-parameter flow cytometry and executing the required compensation. I process tissues necessary for ex vivo assays. I routinely present data not just to my group but to the entire research department. -

Research Associate IiiWyss Institute For Biologically Inspired Engineering Oct 2015 - Feb 2020Boston, Massachusetts, United StatesWhile at the Wyss, I made major contributions to three institute projects. The first was to optimize a coating process for treating various medical surfaces to eliminate blood coagulation and biofouling. While I worked with several different surgical surfaces, most of my efforts were focused on vascular shunts. Once I had optimized the coating parameters, I then supplied these treated shunts for animal studies and further optimized the coating from performance feedback.Another project I worked on was the adaptation of an existing sepsis diagnostic bead ELISA to a commercial 96-well plate format. I optimized the coating of an immunological protein to a polystyrene assay plate and then used the plate to detect pathogen fragments in whole blood. I also optimized the parameters of the assay to dial in the sensitivity and reduce background noise from blood. I then carried out a clinical trial with this plate ELISA on several patient groups from five major hospitals in the Greater Boston area to distinguish trauma patients from infection patients.I also contributed to a vaccine development project by producing plates coated with various antigens. These plates were used to quantify immunological responses from animals in challenge models. -

One of the projects is funded by Hershey's and led to an eventual publication in the journal Clinical Chemistry. The project objective is to examine cognitive effects of theobromine, a compound structurally similar to caffeine. My role specifically is to work with other researchers to develop detection methods to track the compounds in several different types of human samples. The other project is an exploration of using a common shellfish toxin as a local anesthetic. This project also involves optimizing detection methods. LC Mass Spectrometry and HPLC were used during these projects for data collection. I am currently gaining experience with supercritical fluid chromatography. We are using this newer technology to improve our detection methods. In addition, I also run ELISA plate tests for additional lab studies and perform basic tasks to help the lab run smoothly and efficiently. -

I worked with other engineers and scientists to develop superhydrophobic medical surfaces (SLIPS surfaces). My responsibilities also included designing experiments to test the effectiveness of superhydrophobic coatings and analyzing the results. This work gave me great experience with microfluidics, plastics, plasma chemistry, and magnetism. -
Process EngineerRoche Diagnostics May 2012 - Jul 2012Penzberg, GermanyWhile at Roche, I designed and carried out experiments to analyze variables in the fabrication of diagnostic testing chips. In particular, I studied the effects of sample viscosity and temperature on the accuracy of an instrument's pipetting mechanism. I also investigated the significance of several other mechanical variables. At the end of my internship, I delivered a comprehensive report with my results.
