Bioprocess optimisation and development of an adeno-associated viral (AAV) vector platform using stirred-tank bioreactors in Prof Qasim Rafiq lab:- Demonstrated applicability of the Ambr15 high-throughput bioreactor for AAV vector production in suspension HEK293 cells, incorporating design-of-experiment studies.- Received the Excellence in Research Award at the American Society of Gene and Cell Therapy (ASGCT) 2024 annual meeting (major congress with 8,000+ attendees).- Presented research at several conferences/symposiums/webinars.- Collaborating with industry partner Sartorius. Regular meetings to discuss scope and progress of work; invited to present at various company-wide events.

Adeno-associated viral (AAV) vector gene therapy research in Prof Amit Nathwani lab. Projects/research include:- Development of AAV gene therapy for FVII deficiency (haemophilia): molecular cloning of FVII constructs and produced rAAVs to generate protein expression data to determine optimal candidates- Optimised multiplexing platform for qPCR and digital PCR systems to reduce amount of limited clinical sample required- Analysis of microRNA expression from clinical trial samples to identify alternative biomarkers of liver damage in AAV gene therapy- Development of an inducible promoter system to control AAV capsid synthesis and increase full capsid percentage
