Shortly, I have developed a credible in vitro model of the barrier derived from primary mouse choroid plexus epithelial cells suitable for tau transport monitoring and immune cell trafficking. Also, I have studied changes at the barrier at structural (immunohistochemistry) and ultrastructural (electron microscopy studies) level and implemented whole-mount immunostaining and volume imaging (iDisco) to confirm tau localization inside the choroid plexus tissue. Research in the laboratory of Prof. Engelhardt is devoted to understanding the molecular mechanisms involved in the migration of immune cells in the CNS during immunosurveillance and neuroinflammation. They have established in vitro and in vivo (pioneering intravital microscopy of the cervical spinal cord) approaches to study immune cell interactions with the brain barriers in various genetically modified mice lines, mainly in the context of multiple sclerosis and stroke.

My graduate work titled ‘Chaperones linked to ribosomes and cell resistance to environmental stress’ was conducted by the mentoring of Dr. Krisko. We have shown that enhanced expression of the studied chaperones has a positive effect on the robustness of the bacterium E.coli, most likely through enhanced protection of proteins during their synthesis on the ribosome. Bacterium is more resistant to oxidative stress and UV radiation. Protein carbonylation measurements indicated the protection of the protein from oxidative stress by a chaperone.
