Areas of responsibility:In “In vivo Biology”, the primary function is the development of in vivo murine models for allergies and the testing of different treatment concepts. I have experience with immunological, histological, molecular and cellular biological techniques and in vivo experience from mice. As part of a team, I am used to taking responsibility for subtasks as well as my own projects from presentation of the idea, writing the protocol, conducting the experiments, data processing of data, and report writing and presentation. In vitro Cells and serology experience: • Preparation of tissue to single cell level• Preparation of blood and BAL and NAL Lavage for antibody measurement and differential cell counting using sysmex and nucleolar cell counting• In vitro murine Tcells proliferation in culture, ³H-Thymidine radioactive label• Isolation of cells using MS Columns, Leckocytes percoll gradient, Lymphocytes Lympholyte Separation• Staining with cell surface markers (CD) on spleen, lungs, BAL cells • FACS Fortessa analyses• Antibodies and Cytokines assays: Diverse ELISA and Beads immunoassays Molecular Biology experience:• Genotype, DNA from tail tissue, PCR, Agarose gel electrophoresis In vivo experience:• Injecting mice using i.p, s.c, i.n, i.d• Blood sampling from cheek, eye, tail• Collection and preparation of relevant tissues spleen, lungs, lymph nodes• Lung perfusion• BAL, Bronchoalveolar Lavage• NAL, Nasal Lavage

Responsibilities: In vivo experience:• Daily handling of mice: breeding, injection of hormones and earmarking• Operative interventions in mice under microscope: egg retrieval and embryo transfer to other mice Molecular biological experience:• Genotyping using DNA purification from tail biopsies, PCR and slot blot Histological experience:• Organ removal: brain and eyeball• Techniques for analysis of tissue: cutting in cryostat of brain and optic nerve, antibody staining of tissue specimens, protein purification from tissue and western blot

Responsibilities:• Molecular biological experience:• Purification of DNA from human blood and plasmid, agarose gel electrophoresis, PCR, single stranded conformation polymorphism (SSCP), sequencing (with both radioactive and fluorescence labelling), restriction fragment length polymorphism (RFLP), cloning Cell immunological experience:• Transient transfection in NIH 3T3-cells and alpha-TC cells, CAT-assay and electro mobility shift assay (EMSA), western blot
