My PhD, entitled 'Targeting the p53-MDM2 Protein-Protein Interaction', investigated producing novel inhibitors of the MDM2-generated negative feedback loop that degrades the tumour suppressor p53 in its wildtype form, hence forming an important pathway in the pathophysiology of oncology. Inhibitors investigated included a combinatorial library of isoquinolin-1-ones, click-based chlorofusin analogues (that is, the natural product chlorofusin peptide modified to incorporate an azide and conjugated to an aromatic alkyne partner) and analogues of the azaphilone chromophore of the natural product chlorofusin.Compounds were analysed through an in-house fluorescence anisotropy assay followed by an MTS antiproliferative assay and all compounds were fully characterised where possible using physical methods. Additional characterisation was undertaken using saturation transfer difference nuclear magnetic resonance spectroscopy.
