My overall responsibilities include a combination of NMR instrument maintenance and troubleshooting, aiding the medicinal chemists with structure elucidation needs via experimentation setup and interpretation, and innovative project work. The main projects I am responsible for are: management of the NCATS Fragment-Based Screening Program, method development and implementation of our new automation-based qNMR workflow, and analytical processing workflow development using full automation with robotics. All work performed must adhere to NIH/government-level, documentation, confidentiality, and reporting standards. For instrumentation, I am responsible for one 600MHz and four 400 MHz VNMRS instruments equipped with autosamplers, each of which uses a 5 millimeter PFG AutoX dual broadband probe outfitted with a ProTune module allowing for NMR spectra of high-band (1H, 19F) and low-band (13C, 15N, 31P) nuclei. Our 600 MHz magnet is equipped with an XR401 sample cooler attached to the probe, which enables variable temperature experiments ranging from −40°C to 105°C. The robotics instruments I work with for sample preparation and processing include a Sirius Multi-Tasker, Omni-Tasker, and Micro-Tasker. -

My job was to synthesize, purify, and characterize a series of alkaloids for structure-activity relationships (SAR) studies. I performed organic reactions using standard benchtop synthesis, as well as microwave-aided synthesis (Biotage), and most of my purification methods were performed on an Isolera-One Flash Chromatography (Biotage) system. As my time at CUW progressed, I regularly worked on scaling up various reactions (mg to g scales) for non-human primate testing. Analytical instruments and techniques used for characterization included: 1D and 2D nuclear magnetic resonance (NMR) spectroscopy (Varian 500 MHz), high-performance liquid chromatography (HPLC; DionexUltimate 3000 UHPLC and HPLC), Raman spectroscopy (RIGAKU Progeny ResQ 1064), near infrared spectroscopy (NIR; Metrohm NIRSystems DS2500 Analyzer), infrared spectroscopy (IR; Perkin Elmer Spectrum II), and mass spectrometry (MS; AB SCIEX 4000 Q Trap LC/MS/MS). Additionally, I frequently performed custom compound synthesis and HPLC/UPLC method development by request for collaborators. Contract work of note included: synthesis of target compounds for collaborating universities using fully, partially, and/or minimally developed synthetic routes, as provided by the client; designing synthetic route options and creating a proposal with the costs associated with time, equipment use, and materials; HPLC method development for absorption and desorption studies to determine the effectiveness of Deterra Drug Deactivation System per the request of the Milwaukee County Health Department; and developing an HPLC (analytical)/UPLC (preparatory) chiral chromatography method for the separation of a racemic mixture of large quantities of a lead compound. -
