About Rich James

I run an academic research lab interested in uncovering biochemical mechanisms that contribute to drug resistance and to immunological disease. To do so, we investigate many features of protein behavior including post-translational modifications, protein-protein interactions, interaction with drugs and ask how mutation impacts these processes. I am also the co-leader of the B cell engineering program at Seattle Children’s. My lab recently developed a genome engineering technique that can be used to edit primary human B cells, which can subsequently differentiated into antibody secreting cells ex vivo. In addition to studying genetic variants, we also are developing new ways to engineer B cells to express and secrete human proteins with the eventual goal of developing immunotherapies for diseases caused by defects in secreted proteins (e.g. hemophilia).     I directed several high-throughput screens aimed at identifying small molecule and protein regulators of Wnt/beta-catenin signaling. While working on these projects, we established a framework for integrating high-throughput screening and quantitative proteomics data. This work identified several enzymes that inhibit Wnt/beta-catenin signals that could be exploited to ameliorate diseases or phenotypes caused by diminished Wnt/beta-catenin activity.
