About Sean Lund
I use a combination of traditional synthetic chemistry and newly developed synthetic biology approaches to generate a portfolio of natural products and their analogues in a scalable manner. I apply high-throughput screening, protein and metabolic engineering, synthetic strategies, and state-of-the-art biotechnological techniques to efficiently design pathways and routes for small molecule and library production. I am currently leading an enzymology subgroup within a larger project. My group of four is tackling multiple enzymatic bottlenecks using a combination of rational and random protein engineering approaches. I am responsible for interfacing with other subgroups of specialties including HTS, analytics, scientific computing, metabolic engineering, and fermentation in order to propose hypothesis driven experiments and enzymatic limitations that could be addressed through protein engineering.

I am part of the enzymology team at Amyris. I have worked on two separate projects and have gained experience working on a variety of enzyme classes in several different metabolic pathways in yeast. I have gained knowledge in the evaluation of redox balancing and in cosubstrate supply and how they differ based on cultivation conditions. I continue to hone my skills at identification of bottlenecks and evaluation of enzyme engineering opportunities versus metabolic engineering challenges.

My project aims to develop a platform for the fermentation of natural product analogs. The goal of this project is the application of the technology as a biological equivalent to combinatorial chemistry. My experiments have also yielded advances in the knowledge of chemical reaction mechanisms and understanding of the investigated enzymes. I have learned a variety of techniques stretching from organic synthesis to high-throughput screening and enzyme evolution. I am currently a graduate student researcher in the lab of Dr. Gavin Williams and am funded by the Chancellor's Innovation Award at NC State.
