Investigated the mechanism of alternative protein trafficking in the yeast K. lactis by examining the differential use of transcriptional start sites as a B.Sc. thesis project which led to a paid position by the end of 2009. This involved the isolation of yeast RNA followed by reverse transcription and PCR amplification of target transcripts. These were cloned into plasmids and then transformed into bacteria before positive colonies were picked. Plasmid DNA was purified and submitted for sequencing. This led to the identification of multiple transcripts for the target gene (facilitated using BLAST). Differential translation start sites (transcription was investigated during my thesis) were then investigated. A novel knockout strain of K. lactis was generated (by recombination), and this strain was transformed with multiple expression plasmids containing the target gene lacking individual translational start sites. Mutations and restriction sites were introduced by PCR, which allowed for the ligation of multiple fragments into a series of expression plasmids for this study.
