I did QC for machine generated gene sequence by Chromas, and to identify the similarity of sequences all the sequences were submitted to online BLAST (basic local alignment search tool) program at the NCBI website. After reference sequence retrieval from GeneBank database I did multiple sequence alignment by using ClustalW Multiple Alignment in Bioedit. I did phylogenetic analysis by MEGA to rates of molecular evolution, inferring ancestral sequences, and testing evolutionary hypotheses. I also analysis clinical manifestation, prevalence, age distribution and genetic diversity by several Bio statistical software and tools like Microsoft Excel, SPSS, open EPI etc. I found highest prevalence among PWID, HCV genotype was 31% 3a, 23.8% 3b, 4.8% 1b and 2.4% 1a where all 1b genotype show Amino Acid substitution mutation to resist Sofosbuvir drug. Virology laboratory of icddr,b had a research collaboration with Karolinska Institute initiated through a Swedish Research Council Grant on a study that involves transfer of technology relating to genetic analysis of HIV strains collected in Bangladesh. As a part of this collaboration program I Joined a training program on ‘Laboratory assays pertaining to HIV-1 genomics’ at Division of Clinical Microbiology, Department of Laboratory Medicine Karolinska Institute, Stockholm, Sweden.
