I am interested in the intersection of nutrition, aging, and age-related diseases of the visual system, primarily age-related macular degeneration and cataracts. I use model organisms and genetic approaches to understand disease pathobiology and pathetiology with goal of revealing new therapeutic possibilities. My research on AMD is characterizing the roles of dietary glycemic index and genetic factors in development of AMD and therapeutic intervention via dietary change or pharmaceuticals. My research on cataracts is evaluating the function of the ubiquitin proteolysis pathway in development and homeostasis of the lens. Combined, my research will impact over 90% of a continually aging population and offer new interventions in vision-threatening diseases. -

Research FellowBrigham And Women'S Hospital Mar 2004 - Jan 2012My work in the Maas lab centered on two essential pathways required for lens formation. The first is Notch signaling, where I demonstrated essential roles for Notch signaling during lens induction, differentiation, and proliferation. I also identified Notch target genes involved in these processes, several of which may be deregulated during abnormal tissue growth and morphogenesis, such as that during tumor development. The second is elucidating novel upstream regulatory factors involved in Pax6 initiation during lens induction. I identified Prep1 as such an essential upstream factor and used a systems biology and genetic interdisciplinary approach to characterize a new regulatory mechanism whereby Prep1 binds to two lower-affinity binding sites in Pax6 regulatory sequences, which together synergistically activate correct levels of Pax6 expression at the correct time during lens development. -
Postdoctoral FellowHarvard Medical School Sep 2003 - Mar 2004My research in the Cepko lab related to understanding different features of retinal development. With a particular interest in the homeodomain transcription factor Chx10, my work described the upstream transcriptional regulation of Chx10, the function of Chx10 in bipolar cell differentiation, and early functions of Chx10 in maintaining retinal versus retinal pigment cell identity. As a genetic tool, I created a transgenic mouse line that expresses Cre recombinase, green fluorescent protein, and alkaline phosphatase under control of Chx10 regulatory elements. This mouse has been widely used to perform retina-specific gene knockouts and to perform fate mapping experiments in the mouse retina.
