►A research scientist in the Viral Vector Core in National Institute of Environmental Health Sciences (NIEHS/NIH), with a Ph.D. in Microbiology and Immunology. Responsible for virus production, titration, and validation, including lenti/retroviruses, adeno-associated virus (AAVs), Zika virus, Rabies virus, respiratory syncytial virus (RSV) and human immunodeficiency virus (HIV). I am also developing new viral vector tools for gene delivery both in vitro and in vivo. Meanwhile, with prior eight years research experience in Neuropharmacology Group at NIEHS, I have a strong background in investigating the role of the neuroinflammation in mental disorders and neurodegenerative diseases. Specialize in design, develop, and implement experiments in neuron-glia interactions, neuroinflammation, neurodegeneration, and neuro-infectious diseases. Research FellowNational Institute Of Environmental Health Sciences (Niehs) Jun 2012 - Jan 2017Research Triangle Park, Nc, UsI have embarked on several very ambitious projects to study the interaction between microglia and astroglia in inflammation-mediated neuro-degenerative diseases using different primary culture systems, neuro-degenerative rodent models and molecular biology techniques. Despite the fact that glial interaction has been known for some time, the basic mechanism governing this regulation in neuro-degenerative stage remains unclear. The long-term goal is to translate these studies from the bench towards the treatment of neuro-degenerative disorders, including Parkinson's disease.I also collaborate with other laboratories at NIEHS to study the molecular mechanism of how microglia-derived free radicals cause neuronal damage in neuro-degenerative diseases.The following is the summary of my work at this position:• Studied immune regulation of glial cells in the central nervous system by detecting pro-inflammatory cytokine and neurotrophic factor expressions using real-time PCR, ELISA and Western Blot. • Developed and optimized protocols to detect proteins from tissues and from culture supernatant.• Created a state-of-the-art environment for tissue cultures, molecular cloning and protein work in the laboratory, including theories and technologies.• Provided technical advice and supports to solve research problems for the supervisor and other group members.• Reported high-quality and reproducible results to the supervisor on a regular basis.• Attended weekly group meeting for critical data analysis and brainstorming.• Coached and trained postdoctoral fellows and postbac interns.• Researched new methodologies and equipment for the laboratory. Obtained competing quotes and presented cost-benefit analysis to the supervisor.• Supervised the research group to comply with the safety regulations of the institute. -
Visiting FellowNational Institutes Of Health, National Institute Of Environmental Health And Sciences Jun 2010 - Jun 2012Bethesda, Md, UsI performed experiments to study the molecular mechanism of histone deacetylase (HDAC) inhibitor elicited neuroprotective effect. The HDAC inhibitor I used was suberoylanalide hydroxamic acid (SAHA). I found that SAHA could trigger neurotrophic factor (e.g. GDNF and BDNF) released from astroglial cells. SAHA could enhance histone protein hyperacetylated in GDNF promoter region. This study offered a potent therapeutic drug for neurodegenerative disease therapy. The following is the summary of my work at this position:• Studied mechanism of neurotrophic gene regulation in astroglia using Chromatin-Immunoprecipitation (ChIP) assay, Western blot, ELISA, real-time PCR and primary cell culture. • Provided technical advice and supports to solve research problems for the supervisor and other group members.• Reported high-quality and reproducible results to the supervisor on a regular basis.• Attended weekly group meeting for critical data analysis and brainstorming.• Coached and trained postdoctoral fellows and postbac interns.• Researched new methodologies and equipment for the laboratory. I found that virus could use host cell transcription factor to increase viral replication by activating promoters of viral productive cycle genes through binding to its corresponding sequences in the viral promoters.• Proposed a new model of how drug-resistant herpes virus reactivation from infected neurons. • Designed experiments and developed protocols to verify the proposed model, including promoter activity assay, viral gene and protein expressions, ChIP assay, and in vivo animal studies.• Presented results in written and oral formats in seminars, and poster presentation in international conferences. -
