    Project- Explored the therapeutic efficacy of Ruxolitinib (Jak1/2 inhibitor) in the prevention of bone marrow failure and acute myeloid leukemia development in the rare genetic disorder of Fanconi Anemia.About the project: Fanconi Anemia (FA) is a congenital disorder of DNA repair with hematologicmanifestations that may include bone marrow failure (BMF). Previously, we determined that episodes of emergency (stress) granulopoiesis accelerated BMF in Fancc-/- mice; a murine model of FA. Emergency granulopoiesis is the episodic process for granulocyte production during the innate immune response. We found that granulocytosis failed to develop in Fancc-/- mice in response to emergency granulopoiesis stimuli; associated with apoptosis of hematopoietic stem cells and differentiating granulocytes. Fancc-/- mice died with profound anemia during repeated challenges. In the current studies, we found impaired erythropoiesis in Fancc-/- and wild type mice during emergency granulopoiesis, associated with decreased serum erythropoietin and suppression of erythroid progenitor production by IL1β. During steady state resumption, compensatory erythroid progenitor proliferation was associated with DNA-damage and apoptosis in Fancc-/- mice, but not wild type controls. Since Jak2 is activated by the erythropoietin receptor, we investigated the impact of a Jak1/2 inhibitor (Ruxolitinib) on this process. During repeated episodes of emergency granulopoiesis, we found Ruxolitinib-treatment prolonged survival and decreased anemia in Fancc-/- mice. Ruxolitinib is chronically administered for various clinical indications, suggesting potential feasibility of using this agent for bone marrow protection in FA.Mentor- Dr. Elizabeth Eklund
