Finally, determination of Enzyme assay using Kinetics editor on Nanodrop Spectrophotometer as well as performing Bradford protein assay (binding of Coomassie Brilliant Blue G-250 to purified protein). Then the results are analyzed by Protein purification table to calculate Total milligram of protein , total enzyme activity of both crude and dialyzed extract, specific activity, Fold purification and %yield of protein.2) Cibacron Blue-Agarose chromatography- Performing column chromatography (step gradient) for further of purification of dialyzed Lactate Dehydrogenase enzyme using a stationary phase specific for proteins having 'Rossmann fold'.Then by performing the enzyme activity and Bradford assays on the pooled fractions of chromatography students were successful in achieving high fold purification and high %yield of Lactate Dehydrogenase enzyme.3) High-performance Liquid chromatography(HPLC)-HPLC separation of steroid hormones by exploring the varying effects of solvent percentage on the eluting power; in the process to discover the optimal solvent mixture which allows the baseline separation four steroid peaks within a minimum of time on C-18 reverse phase column. Finally, students were successful in identifying, determining the retention times all four hormones given in the sample.4) Purification of a Maltose Binding protein(MBP)-Paramyosin Fusion protein by Affinity chromatography. SDS-PAGE and Western Blot on purified fusion protein.5) Two-dimensional Gel electrophoresis of protein mixtures.
