About Simanga R. Gama, Ph.D.

 I am a self-driven researcher who is enthusiastic about studying proteins/enzymes and their role in disease, and more especially as therapeutic targets. In my current position, I study protein-protein, protein-DNA and protein-small molecules interactions using biochemical and biophysical tools. During my previous position as a postdoctoral fellow, I looked at enzymatic pathways of phosphonate catabolism with emphasis on glyphosate degradation. This sparked an interest in how enzymes can be used as bioremediation or bio-restoration tools following chemical pollution of the environment. As a PhD candidate, I characterized KDO8P synthase from Campylobacter jejuni. KDO8P synthases are enzymes found in all Gram-negative bacteria and are antimicrobial targets. We developed a slow-tight-binding inhibitor of KDO8P synthase which afforded us the opportunity to learn more about developing effective inhibitors for multi-subunit enzymes. My cumulative technical experience includes protein-protein, protein-DNA/small molecule interactions (Microscale thermophoresis, TR-FRET, ELISA, DSF, x-ray crystallography, ITC), enzyme kinetics & inhibition, UV spectrophotometry, protein expression and purification, NMR, HPLC, FPLC, radioisotope labeling. I also have experience in molecular biology techniques such as, cloning, site-directed mutagenesis, PCR.  I can critically analyze data from my own research in comparison with literature findings, communicate my results through scientific writing and oral presentation (see publication section). I also have (and growing) collaborative experience across colleagues, collaborators, and clients. I have leadership and in-lab-teaching experience obtained as a PhD candidate, Postdoctoral fellow and currently as a Scientist.
