During the past five years I have been investigating about the glycoprotein quality control system in the Endoplasmic Reticulum (ER). Several aspect of the glycoprotein folding in the ER are still unknown, and my research has been focused on the design of model glycoprotein able to form correctly folded and misfolded species in a controlled manner to investigate the interaction with ER quality control system enzymes. Recently I have been studying the preparation of Sep15, an ER resident selenocysteine protein, which seems to have an oxido-reductase function, but which role it is still not defined. Such protein has been already prepared in the past, but using cysteine in place of the selenocysteine, which we believe is essential to clearly understand its function.

During the JSPS research fellowship in Japan my work was based on the application of microwave irradiation to improve the synthesis of Gold Colloidal NanoParticles (GCNPs). Following the report of the application of the GCNPs as solid support for solid phase carbohydrate synthesis the ultimate goal of the research theme was to develop and improve the synthesis of the GCNPs and their application in order to build a versatile and solid water-soluble platform to be used for carbohydrate solid phase synthesis, enzymatic reaction, and direct biological assay.

My Ph.D. research focused on application of “click chemistry” to modify carbohydrates. The project was mainly dedicated to the synthesis of reversible and irreversible glycosidase inhibitors. The main targets of the reversible inhibitors were the different glycosidases involved in the starch metabolism in the barley seeds. The irreversible glycosidase inhibitors were envisioned to work as tools for understanding glycosidases mechanism or to be used for post translational modifications of proteins. In addition to the main theme, I have been also working on the enlargement of the sugar chain of small potential glycosidase inhibitors by using chemo-enzymatic synthesis, in order to target enzyme with extended binding site (i.e. β-glycosidase) and on synthesis of pseudo-starch fragments by Cu (I)-catalyzed Huisgen’s 1,3-dipolar cycloaddition (“click” chemistry).
