Scientist I, Synthorx 2018-Present  Played a pivotal role in identifying lead AI candidate using SAR, under highly aggressive project timelines. Purified and characterized mg amounts of site-specific bio-conjugated proteins under GLP conditions for IND enabling, pre-clinical studies. Prepared biologics for ELISA and interpreted dose-response curves. Wrote reports, oversaw SOPs and batch records and issued COAs for purified biomolecules. Contributed to platform development by carrying out high-throughput fluorescence-based enzyme solubility/activity screen.

Projects include:• Laid the structural and biochemical foundation for the selective recruitment and deubiquitination of substrates at the yeast rDNA locus (published in Genes and Development, 2017). • Evaluating bacterial, yeast and insect cells as models for expression of meiotic proteins for structural studies.• Using ligand and structural mimetics, developed an atomic understanding of how a monomethyl reader is recruited to the nucleosomes• Investigating the molecular mechanism of sister meiotic chromosome co-segregation using crystallography resulting in 3 co-crystal structures and biochemical characterization using ITC, FP, MALS and in-vitro pulldowns.• Obtained mg amounts of a previously recalcitrant protein from E.Coli after screening >400 single and dual expression constructs to set crystal trays and paved the way for biochemical characterization.• >400 single and dual expression constructs screened for protein expression. Painstaking work led to isolation,purification and initial crystal screening of sub-complexes from bacteria, something that has thus far eluded the field.• Molecular biology techniques like cloning (ligation dependent and independent including LIC, CLIC, Isothermal/Gibson assembly), pulldown and immunoprecipitation experiments to assess protein-proteininteractions.
